加味小青龙汤及其拆方对变应性鼻炎大鼠鼻黏膜病理形态和IL-4含量的影响-《环球中医药》

摘要:: 目的观察加味小青龙汤及其拆方对变应性鼻炎大鼠鼻黏膜病理形态和血浆白细胞介素4(IL-4)含量的影响,探讨该方益气温阳化饮的作用机制及其组方配伍规律。方法将90只大鼠随机分成正常对照组、模型组、益气组、温阳组、化饮组、益气温阳组、益气化饮组、温阳化饮组和加味小青龙汤组,每组10只。采用抗原佐剂全身致敏与局部攻击方法,制备大鼠变应性鼻炎模型,正常对照组和模型组灌服生理盐水,其它各治疗组给予相应的药物,每天一次灌胃给药,连续4周。检测加味小青龙汤及其拆方对变应性鼻炎大鼠鼻黏膜病理形态和血浆IL-4含量的影响。结果模型组可见鼻黏膜水肿、充血,上皮不完整,重度炎细胞浸润,嗜酸粒细胞浸润明显,黏膜上皮坏死。各治疗组炎症浸润程度均有改善,以加味小青龙汤组效果最好。模型组血浆IL-4含量明显升高,加味小青龙汤治疗组及其拆方组血浆IL-4含量明显降低,与模型组比较有统计学意义(P<0.05或P<0.01),其中以加味小青龙汤组与各拆方组比较有统计学意义(P<0.05)。结论加味小青龙汤及其拆方各组药物,均具有不同程度的改善鼻黏膜病理炎症浸润程度、降低血浆IL-4含量的作用,加味小青龙汤组的疗效最好,体现了温阳、益气、化饮的合理配伍表现出协同增效的整体调节作用,揭示了加味小青龙汤配伍的高度科学内涵。

Abstract:: ObjectiveTo observe the added Xiaoqinglong Decoction and its ingredientreduced prescription on pathological nasal allergic rhinitis in rats and plasma levels of　interleukin4(IL4), and to explore the mechanism of tonifying qi and warming yang to expel retained morbid fluid and their combination compatibility of the law. MethodsThe 90 rats were randomly divided into nine groups, namely, normal control group, model group, reforcing qi group,warming yang group, expelling retained morbid fluid group, reforcing qi and warming yang group, tonifying qi and expelling retained morbid fluid group, warming yang to expel retained morbid fluid group, the added Xiaoqinglong Decoction group,10 rats in each group. Rat model of allergic rhinitis was made by the use of adjuvant systemic antigen sensitization and local attack, the normal control group and model group were fed with normal saline, the other group was treated with appropriate drugs, once a day oral administration, continuous for 4 weeks. The pathological nasal allergic rhinitis and plasma IL4 level in rats were tested. ResultsThe control group showed that nasal was swelling, congestion, epithelial incomplete, severe inflammatory cell infiltration, eosinophil infiltration, epithelial necrosis. Inflammatory infiltration in all treatment groups had improved and the added Xiaoqinglong Decoction was best. Compared with control group, Plasma IL4 Content was significant lower in the added Xiaoqinglong Decoction and its ingredientreduced prescription group(P<0.05 or P<0.01). While compared with other its ingredientreduced prescription groups, there was statistical significance in the added Xiaoqinglong Decoction group (P<0.05). ConclusionThe added Xiaoqinglong Decoction and its ingredientreduced prescription can improve inflammatory infiltration of nasal pathological and regulate plasma levels of IL4.The added Xiaoqinglong Decoction demonstrated best effects, which implied that the compatibility of the overall regulatory role of synergies of tonifying qi and warming yang to expel retained morbid fluid and a high degree of compatibility of scientific content.
