摘要:
目的研究参附芎泽方药对阿霉素所致心力衰竭大鼠心功能的影响,并探讨其作用机制。方法 SD大鼠随机分为2组:阿霉素组40只,腹腔注射阿霉素,2.5 mg/kg,3次/周,连续2周; 生理盐水组,10只,腹腔注射同剂量的生理盐水,连续2周,随后正常饮食4周。2周后阿霉素组死亡7只,余下33只随机分为4组:阿霉素对照组9只,正常饮食; 低剂量中药组(每毫升煎液含生药0.5 g)、中剂量中药组(每毫升煎液含生药1 g)、高剂量中药组(每毫升煎液含生药2 g),每组8只,持续灌胃4周,剂量均为12 ml/(kg.d),4周后测量每组大鼠体质量、心脏质量,计算心脏质量/体质量; 右侧颈总动脉插管至左心室,采用多导生理信号采集处理系统监测大鼠血流动力学指标; 病理切片,HE染色,光镜观察左心室心肌组织变化。结果阿霉素组相对于生理盐水组大鼠体质量、心脏质量及心脏质量/体质量均降低,且体质量的降低更为显著(P<0.05); 血流动力学指标:平均动脉压、心率、左心室收缩压、左心室内压最大上升及下降速率均下降,左心室舒张末期内压升高(P<0.05); 镜下观察左心室心肌组织损伤明显,经各方药组灌胃干预4周后,以上各项指标均有所改善,且以大剂量方药组的作用显著。结论参附芎泽方药对阿霉素所致心衰具有较好的防治作用,能够明显改善心衰大鼠的心脏功能。
Abstract:
ObjectiveThe present study was carried out to investigate the effects of Shenfu Xiongze Decoction on cardiac function of adriamycininduced heart failure rats. MethodsSD rats were randomly divided into 2 groups: adriamycin (ADR) group (n=40): 40 animals received ADR (2.5 mg/kg,i.p.) three times a week,for 2 weeks; control group (n=10),animals received equivalent volume of saline (i.p.). After 2 weeks,7 animals died. 9 animals act on ADR control;8 animals each received Shenfu Xiongze Decoction (low dose:0.5 g crude drug in 1ml physic liquor)for 4 weeks;8 animals each received Shenfu Xiongze Decoction (moderate dose:1 g crude drug in 1 ml physic liquor) for 4 weeks;8 animals each received Shenfu Xiongze Decoction (high dose:2 g crude drug in 1ml physic liquor) for 4 weeks. The ADRinduced heart failure and protective action of Shenfu Xiongze Decoction were determined and quantitated with the use of body weight (BW),heart weight (HW),hemodynamic measurements and histopathological procedure. ResultsBW,HW,mean arterial pressure(MAP),heart rate(HR),left ventricular systolic pressure (LVSP) and maximal rising and falling rate of left intraventricular pressure (±LVdp/dtmax) were decreased,while left ventricular enddiastolic pressure (LVEDP) inscreased and histopathology revealed myocardial lesions indicative of ADR induced heart failure. In contrast,administration of Shenfu Xiongze Decoction significantly attenuated these effects. ConclusionShenfu Xiongze Decoction is potentially protective against ADRinduced heart failure and improve cardiac function.
