摘要:
目的探讨绿原酸(CGA)对阻塞性黄疸大鼠肝纤维化的治疗作用及其机制。方法将72只SD雄性大鼠随机分为对照组(SO组),胆总管结扎+生理盐水组(BDL+NS组),胆总管结扎+CGA组(BDL+CGA组)。分别于术后7、14和21天用RT-PCR法和Westernblot法检测bcl-2、Bax在mRNA水平和蛋白水平的表达。用RT-PCR法检测I、III型胶原mRNA的表达。结果与BDL+NS组比较,胆总管结扎7天、14天时,BDL+CGA组可以提高Bax/bcl-2的比值,减少I、III型胶原RNA的表达。结论 CGA在大鼠阻塞性黄疸早、中期有促进HSC凋亡作用,减轻了阻黄时肝的纤维化,减轻了阻黄时肝的损伤。
Abstract:
Objective To investigate the effects and mechanism of chlorogenic acid on fibrosis of liver in the rat with obstructive jaundice. Methods Seventy-two male SD rats were randomly divided into three groups, sham operation group(SO), bile duct-ligation+normal soldium group(BDL+NS)and bileduct-ligation+ chlorogenic acid group(BDL+CGA). The mRNA expression of collagen I, collagen III, bcl-2 and Bax as well as the protein expression of bcl-2 and Bax were examined on days 7,14,21 after operation. Results On the 7th, 14th day, the protein expression of Bax/ bcl-2 was increased in BDL+CGA group compared with BDL+NS group.The mRNA expression of collagen I and III in BDL+CGA group was decreased compared with BDL+NS group. Conclusion CGA has a protective effect on fibrosis of liver in the initial,metaphase stage of obstructive jaundice by promoting apoptosis of hepatic stellate cell.
