摘要:
目的观察黄连碱对Sprague Dawley大鼠关节软骨细胞体外增殖及转化生长因子β1mRNA基因表达的影响,探讨其治疗骨关节炎的作用及可能机制。方法取SD大鼠关节软骨,剪碎后,采用酶消化法原代分离培养软骨细胞,取处于对数生长期第3代细胞随机分组,实验组加入含10 μm黄连碱的培养基,对照组仅加入普通培养基。培养48小时后在倒置显微镜下观察软骨细胞形态,MTT法检测黄连碱在1,3,5,7天对软骨细胞增殖影响,实时荧光定量PCR分别检测培养7天后实验组和对照组细胞转化生长因子β1mRNA表达。结果培养48小时后实验组软骨细胞生长密集程度明显优于对照组;MTT检测也显示实验组细胞增殖情况优于对照组;培养7天后实验组转化生长因子β1mRNA表达显著高于对照组。结论黄连碱能促大鼠软骨细胞增殖,并提高转化生长因子β1mRNA的表达,提示这可能是其治疗骨关节炎的可能机制之一。
Abstract:
ObjectiveTo observe the effect of coptisine on the proliferation of rat articularchondrocyte and transforming growth factorβ1 mRNA expression, and then to explore the role and possible mechanism of coptisine in the treatment of osteoarthritis. MethodsKnee cartilage was shredded after harvested from SD rats under sterile conditions, and chondrocytes were isolated and cultured by the way of enzymatic digestion. After identifying by toluidine blue staining, the thirdpassage cells in the logarithmic growth phase were cultured in vitro and randomly divided into two groups after adherence. The experimental groups were cultured in DMEM with 10 μm coptisine, while the control group was given with normal medium alone. Chondrocytes morphology was observed under an microscope after 48 hours. 1, 3, 5, 7 days later, Methyl Thiazolyl Tetrazolium (MTT) assay method was adopted to observe the influence of coptisine on the proliferation of chondrocytes.Transforming growth factorβ1 mRNA levels were detected and analysed by quantitative real time polymerase chain reaction(qRTPCR) at 7days after cell culture.ResultsThe cell growth intensity in the coculture group was significantly better than in the control group observed under microscopeafter 48 hours after culture. MTT assay showed that cell proliferation in experimental group were higher than in the control group after 3 days of cell culture.transforming growth factorβ1 mRNA level in the coculture group weresignificantly better than the control group after 7 days of cell culture. Conclusioncoptisine can promote proliferation of chondrocyte and transforming growth factorβ1 mRNA expression, and these may be one of the possible mechanisms thatcoptisine can work in the treatment of osteoarthritis.
