摘要:
目的 观察亚急性毒理实验中不同浓度艾烟对大鼠血清瘦素、肺、膈肌和趾长伸肌腱的影响,为艾烟的安全性评价提供参考。方法 将40只雌雄Wistar大鼠随机分为四组,每组10只,雌雄各半,分别暴露于0 mg/m3、2.5~3.5 mg/m3、8.0~10.0 mg/m3、27.0~29.0 mg/m3的艾烟浓度,200 min/day,5次/周,12周后取材,采用酶联免疫吸附双抗体夹心法(enzyme linked immunosorben tassays,ELISA)检测不同浓度组大鼠血清瘦素、膈肌、趾长伸肌腱肿瘤坏死因子-α(tumor necrosis factor,TNF-ɑ)、白介素- 8(interleukin- 8,IL- 8)的含量,并进行肺和骨骼肌的组织病理学检查。结果 低、中、高浓度组血清瘦素、膈肌和趾长伸肌腱TNF-ɑ、IL- 8含量与空白对照组无显著差异(P>0.05),各组肺、膈肌和趾长伸肌腱病理未见显著改变。结论 亚急性毒理实验低、中、高浓度艾烟对大鼠肺、骨骼肌未见明显损伤。
Abstract:
Objective To investigate the subacute toxicities of moxa smoke on the serum leptin, lung, diaphragm and extensor digitorum longus tendon in rats, so as to provide reference for the safety evaluation of moxa smoke. Methods 20 male and 20 female Wistar rats were randomly divided into four groups(n=10). The experimental groups were respectively exposed to 0 mg/m3, 2.5~3.5 mg/m3, 8.0~10.0 mg/m3 and 27.0~29.0 mg/m3 concentrations of moxa smoke, 200 min/day, 5 times/week. The rats were sacrificed after 12 weeks. ELISA was used to detect the levels of serum leptin, and TNF-α and IL- 8 of the diaphragm and extensor digitorum longus tendon. Histopathological examination was also performed on the lung, diaphragm and extensor digitorum longus tendon.Results No significant difference was observed in the levels of serum leptin, and TNF-α and IL- 8 of the diaphragm and extensor digitorum longus tendon between the control group and experimental groups(P>0.05). Histopathological examination of the lung, diaphragm and extensor digitorum longus tendon did not present significant changes either.Conclusion The subacute toxicity study shows that low, medium and high concentrations of moxa smoke does not cause obvious damage to rats' lung and skeletal muscles.
