摘要:
目的 研究平肺复方对人肺腺癌A549细胞增殖及过氧化物酶体增长因子活化受体(peroxisome proliferators-activated receptor, PPAR)-γ信号通路的影响。方法 运用细胞计数试剂盒(cell counting kit- 8,CCK- 8)比色法检测平肺复方对人肺腺癌A549细胞增殖的影响,并添加PPAR-γ特异性阻断剂GW9662,观察PPAR-γ通路阻断后平肺复方对A549细胞生长的影响。结果 平肺复方生药5 mg/ml和10 mg/ml在作用24小时、48小时和72小时表现出对A549细胞增殖的抑制作用(P<0.05); 单纯加入GW9662对细胞的生长并未产生影响; 加入GW9662的平肺复方仍可对细胞生长产生抑制作用(P<0.05),但这种抑制作用明显弱于平肺复方(P<0.05)。结论 平肺复方的抗肿瘤作用机制可能部分涉及到PPAR-γ信号通路,但并不完全是通过这一条信号通路在发挥作用。
Abstract:
Objective To investigate the growth effect on human lung adenocarcinoma cell line A549 by Pingfei Compound, and the effect on PPAR-γ Signal Pathway induced by it. Methods Proliferation of A549 cells after exposure to the apozem was measured by CCK-8 assay. The PPAR-γ specificity blocker GW9662 was added. Observe if the Pingfei Compound could influence the growth of the A549 cells or not. Results The 5 mg/mL and 10 mg/mL groups presented inhibitive effect on A549 growth at 24h, 48h and 72h time points(P<0.05). GW9662 did not make any difference on cell growth,while the Pingfei Compound supplemented with GW966w showed inhibitory effect on cell growth(P<0.05). Conclusion The mechanism of anti-tumor effect of Pingfei Compound may partly involved in the PPAR-γ signal pathway. But other signal pathways may meanwhile function in the anti-tumor process.
