摘要:
目的研究梓醇对缺血细胞的保护作用机制。方法通过连二亚硫酸钠消除培养基中的氧,在37℃、5% CO2培养箱中,模拟缺血环境中的细胞损伤;再对对照组、模型组、梓醇低、中、高浓度组分别进行细胞存活率、总抗氧化能力(total antioxidant capacity,TAC)、超氧化物歧化酶(superoxide dismutase,SOD)、乳酸脱氢酶(lactate dehydrogenase,LDH)测定。结果梓醇能显著提高细胞存活率,且在5~20 μmol/L呈剂量依赖性,与模型组比较,差异有统计学意义(P<001);梓醇能显著提高细胞培养液上清液中的TAC、SOD,降低LDH水平,且在5~20 μmol/L呈剂量依赖性,与模型组比较,P<001,有统计意义。结论梓醇可以通过抗氧化损伤保护缺糖缺氧细胞。
Abstract:
ObjectiveTo study the protective mechanism of catalpol onchemic cells. MethodsBy sodium dithionite and eliminate medium oxygen at 37℃, 5% CO2 incubator, cell damage of simulated ischemia environment. The cell survival rate, total antioxidant capacity (totalantioxidant capacity, TAC), superoxide dismutase (superoxide dismutase SOD) and lactate dehydrogenase (lactate, dehydrogenase, LDH) determination were compared among the control group, model group, low catalpol group ,medium catalpol group and high catalpol group. ResultsCompared with model group, catalpol can significantly improve the survival rate of cells in a dosedependent manner(5~20 mol/L, P<001). Compared with the model group, catalpol can significantly improve the cell culture supernatant of TAC, SOD, reduce the LDH level with the same trend of survival rate of cells. ConclusionCatalpol can protect OGD cell oxidative damage.
