摘要:
目的 通过动物实验,研究中药复方黛黄芍药散在抑制微血管血栓形成、改善肠道微循环方面对放射性直肠炎的作用机制。方法 选用雌性Wistar大鼠60只,随机分为6组:空白对照组、模型组、地塞米松组及黛黄芍药散高、中、低剂量组,用60Co-γ射线单次盆腔照射制作大鼠放射性直肠炎模型,各组给予相应药物干预,在照射后第14天给药12小时后处死大鼠,取直肠进行免疫组化检测其环氧化酶2(cyclooxygenase-2,COX-2)的表达,采用酶联免疫吸附试验检测各组血清中血栓素B2(thromboxane B2,TXB2)、6-酮-前列腺素(6-keto-prostaglandin F,6-Keto-PGF)的浓度,并计算TXB2/6-Keto-PGF比值。结果 照射后第14天,模型组大鼠COX-2、TXB2含量及TXB2/6-Keto-PGF比值明显升高,而黛黄芍药散各剂量组中上述指标明显低于模型组(P<0.01)。结论 黛黄芍药散可有效抑制放射性直肠炎大鼠肠道微血管血栓生成,改善微循环,其作用机制可能是通过控制COX-2的表达,从而减少TXB2的合成与释放,进而调节TXB2/6-Keto-PGF比值。
Abstract:
Objective To explore the mechanism of Daihuang Shaoyao powder on inhibiting thrombosis and improving the intestinal microcirculation in the rat model of radiation proctitis. Methods Sixty Wistar female rats were randomly divided into 6 groups, normal group, model group, dexamethasone group, high, mediumand, low dosage of Daihuang Shaoyao powder groups. Rat models of radiation-induced proctitis were produced by 60Co-γ ray radiation. The levels of COX-2, TXB2, 6-Keto-PGFand TXB2/6-Keto-PGFwere detected after drugs intervention for 14 days. Results Compared with the model group, the levels of COX-2, TXB2 and TXB2/6-Keto-PGF were obviously decreased in high dosage of Daihuang Shaoyao powder(P<0.01). Compared with the model group, Daihuang Shaoyao powder groups could effectively reduce the level of COX-2,TXB2 and TXB2/6-Keto-PGF(P<0.01). Conclusion Daihuang Shaoyao powder could effectively inhibit the formation of intestinal microvascular thrombosis, improve microcirculation, and the mechanism of action may be through controlling the expression of COX-2, which can reduce the synthesis and release of TXB2, and then adjust the ratio of the TXB2/6-Keto-PGF1 α.
