摘要:
目的探讨补肾解毒活血法对骨髓抑制小鼠造血功能的影响。方法取昆明小鼠60只,随机分成空白对照组、模型组和中药治疗组,每组各20只,模型组和中药治疗组腹腔注射环磷酰胺100mg/(kg?d),连续3天; 造模完成后次日起,中药治疗组给予补肾解毒活血中药浓煎液灌胃,0.36g/(kg?d),空白对照组与模型组灌服等量生理盐水,连续10天。各组随机抽取10只,于造模前1天及造模后第1、3、5、7、10、14天检测小鼠外周血象; 其余10只于造模后第11天,颈椎脱臼处死,检测骨髓造血祖细胞集落形成和骨髓有核细胞(Bone Marrow Nucleated Cell,BMNC)的情况。结果外周血白细胞,模型组和中药治疗组均在造模后第1天降到最低点,与空白对照组相比差异均有显著性意义(P<0.01); 模型组和中药治疗组均在第3天开始回升,中药治疗组在第5天恢复正常且保持稳定,而模型组在第7天出现异常性增高后又逐渐下降至正常。外周血血小板,模型组在模后第5天降到最低然后回升至第10天恢复正常,而中药治疗组自第3天即开始回升,第7天时即与空白对照组持平,其最低值明显高于模型组(P<0.01)。造模后第11天检测粒细胞-巨噬细胞集落生成单位(CFU-GM)、定向体外红系集落形成单位(CFU-E)、红细胞爆裂型集落生成单位(BFU-E)和小鼠骨髓有核细胞数(BMNC),模型组和中药治疗组均低于空白对照组(P<0.05),而中药治疗组高于模型组(P<0.01)。结论补肾解毒活血法治疗骨髓抑制可通过增加骨髓造血祖细胞数量,从而促进环磷酰胺致骨髓抑制小鼠外周血白细胞、血小板的稳定恢复。
Abstract:
ObjectiveTo investigate the effects of Bushen Jiedu Huoxue Recipe on the bloodproducing function in mice of myelosuppression induced by cyclophosphamide Methods60 mice were randomly divided into 3 groups,20 in each group:the blank control ,the model control and the Bushen Jiedu Huoxue group,the latter two groups of mice were injected cyclophosphamide intraperitoneally for 3 consecutive days at 100 mg/kg dailyThe next day after the model was established,the Bushen Jiedu Huoxue group of mice were administrated by gavage with Bushen Jiedu Huoxue decoction for 10 days at 036 g/(kg·d),the same volume of physiological saline was given to the other two groups respectively Thereafter,the peripheral blood was detected on 0、1、3、5、7、10、14 days after model established ;BMNC、hematopoietic progenitor cell colony forming were detected on the 11th day ResultThe WBC of the model group and the Bushen Jiedu Huoxue group descended to the lowest point after 1d the model was established,which had obvious significance compared to the blank control group(P<001),WBC of the model group and Bushen Jiedu Huoxue group both upswung on the 3rd day,while the Bushen Jiedu Huoxue group put back on the 5th day and kept steady,the model group showed up an abnormal raise on the 7th day and then returned to normal The PLT of the model group descended to the lowest point after 5days the model was established and upswung to normal on the 10th day,while the BushenJieduHuoxue group upswung from the 3rd day and recovered to normal on the 7th dayThe lowest point of PLT of the mice in Bushen Jiedu Huoxue group was higher than that of the model group(P<001)The BMNC、CFUGM、CFUE and BFUE of Bushen Jiedu Huoxue group was higher than the model group and lower than the control group(P<001) ConclusionBushen Jiedu Huoxue Recipe may play a key role in increasing the number of hemopoietic progenitor cell of marrow depressed mice induced by cyclophosphamide and enhanced the steady recovery of peripheral blood of WBC and PLT
