摘要:
目的探讨心之络病"孙络绌急"大鼠模型下心肌组织病理变化特征。方法将30只雄性SD大鼠随机分为正常组和造模5分钟组、10分钟组、20分钟组、30分钟组5组,每组6只。采用大鼠股静脉注射垂体后叶素方法制备心之络病"孙络绌急"模型,正常组注射等量生理盐水。采用HE染色方法观察心肌组织病理改变,Nagar-Olsen染色观察心肌组织损伤程度,透射电镜技术观察心肌细胞超微结构的变化。结果光镜下心肌组织HE染色切片在各模型组均未见明显的形态学改变。Nagar-Olsen染色后,各模型组心肌组织均可见到缺血红染的阳性反应区域,并尤以10、20分钟组最为明显。透射电镜下,各模型组均可见心肌细胞凋亡;线粒体嵴排列紊乱、模糊断裂;微血管内皮细胞凋亡,吞饮小泡数量显著增加等超微结构改变。结论心之络病"孙络绌急"大鼠模型初期即可出现心肌超微结构损伤并由此呈现出明显心肌缺血现象。
Abstract:
ObjectiveTo investigate the pathological change of myocardium in constriction of Sun collaterals model. Methods A total of 30 male SpragueDawley rats were random divided into the Control group; the 5min group; the 10min group; the 20min group; the 30min group.Constriction of Sun collaterals model was made in rats by injecting pituitrin through femoral vein.The changes of pathological in myocardium were observed by HE staining,the degree of myocardial ischemic injury in rat was observed by NagarOlsen staining,and the changes of ultra microstructure in myocardium were detected by transmission electron microscope. ResultsThere were no obvious pathology changes by HE staining under the light microscope in all the model groups.The positive regions for NagarOlsen staining were observed in all model groups,especially in the 10min and 20min group.Under the transmission electron microscope,lots of ultra microstructure changes,such as myocardium cell apoptosis,mitochondria ridge arranged disorderly and crest break,microvascular endothelial cell apoptosis,and pinocytotic vesicle significantly increase were observed in all the model groups. ConclusionsEarly myocardial ultra microstructural damage can be induced by constriction of Sun collaterals.
