摘要:
目的 观察糖痹康对糖尿病大鼠坐骨神经II相代谢酶谷氨酸半胱氨酸连接酶亚基(glutamate cysteine ligase catalytic subunit,GCLc)和谷胱甘肽 S 转移酶(glutathione s transferase,GST) pi蛋白与mRNA的影响,探讨糖痹康对糖尿病周围神经病变大鼠坐骨神经保护作用机制。方法 雄性SD大鼠,高脂饲料与小剂量链脲佐菌素(streptozotocin,STZ)联合诱发糖尿病大鼠模型,模型成功后将糖尿病大鼠随机分为5组:模型组、α 硫辛酸(alpha lipoic acid ,ALA)组(0.0268g/(kg·d),糖痹康高(2.5g/kg)、中(1.25g/kg)、低(0.625g/kg)剂量组,每组10只,另将雄性SD大鼠10只设为正常组。治疗12周后测大鼠右侧坐骨神经传导速度;Western blot检测大鼠坐骨神经中GCLc和GST pi蛋白的表达;实时荧光定量PCR检测大鼠坐骨神经中GCLc和GST pi mRNA的表达。结果 与模型组相比,12周后ALA组、糖痹康高、中剂量组大鼠坐骨神经传导速度明显升高(P<0.05);ALA组、糖痹康高、中剂量组GCLc和GST pi蛋白及mRNA表达均明显高于模型组(P<0.05)。结论 糖痹康可通过诱导Ⅱ相代谢酶GCLc和GST pi改善糖尿病大鼠坐骨神经损伤。
Abstract:
Objective To observe the effect of Tangbikang(TBK) on the protein and mRNA of Phase II metabolizing enzymes of GCLc and GST pi of sciatic nerve in diabetic rats to investigate the protection of the sciatic nerve in rats with diabetic peripheral neuropathy. Methods Male SD rats were selected for research,high fat diet combined with low dose of streptozotocin(STZ)to induced diabetic model, and the rats were randomly divided into 5 groups:model group,alpha lipoic acid (ALA) group,TBK high dose group(2.5g/kg),medium dose group(1.25g/kg)and low dose group(0.625g/kg),10 rats in each group and another 10 male SD rats were put into normal group. After 12 weeks treatment,the right sciatic nerve conduction velocity was measured by direct method,and the protein and mRNA of GST pi and GCLc were detected by Western blot and real time PCR. Results [WTBZ]After 12 weeks,the sciatic nerve conduction velocity in the ALA group,TBK high and medium dose groups was significantly higher than that in the model group(P<0.05). The expression of GST pi and GCLc protein and mRNA in the ALA group, TBK high and medium dose groups were significantly higher than those in the model group. Conclusion [WTBZ]TBK can improve the sciatic nerve injury of DPN rats by inducing Phase II metabolizing enzymes of GCLc and GST pi of sciatic nerve in diabetic rats.
