目的 基于肝肾同源理论,观察糖尿病肾病(kk-ay)小鼠后期谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶含量(aspartate transaminase,AST)变化以及芪水煎对肝、肾基质金属蛋白酶-9(Matrix metalloe proteinas-9,MMP-9)及基质金属蛋白酶组织抑制因子-1(tissue inhibitor of metalloproteinase-1,TIMP-1)表达的影响。 方法 采用kk-ay小鼠高糖高脂饮食4周建立糖尿病肾病模型后,随机分为模型组、黄芪水组、通心络组,设立C57BL/6J小鼠作为对照组。予芪水煎及通心络干预12周后,监测各组小鼠血ALT、AST,采用Western Blot、Realtime-PCR检测肝、肾组织MMP-9和TIMP-1蛋白含量及mRNA表达水平。 结果 与模型组比较,芪水煎组AST显著降低(P<0.01)、ALT呈下降趋势(P>0.05),肝MMP-9蛋白及mRNA表达显著升高(P<0.01),TIMP-1蛋白显著降低(P<0.01),mRNA表达降低(P<0.05),肾MMP-9蛋白及mRNA表达升高(P<0.05),TIMP-1蛋白及mRNA表达降低(P<0.05)。结论 kk-ay小鼠后期会出现肝损伤,芪水煎对kk-ay小鼠肝、肾损伤具有一定的保护作用。
Abstract:
Objective To investigate the content changes of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)of diabetic nephropathy(kk-ay)mice in later stage, and to investigate the influence of Qishui decoction on the expression of MMP-9, TIMP-1 in the hepatic and nephritic tissues based on the theory of “homogeny of liver and kidney”. Methods The diabetic nephropathy model was established by kk-ay mice with high glucose and high fat diet for 4 weeks. The mice were randomly divided into model group, Qishui group and Tongxinluo group, and C57BL/6J mice were used as control group. After 12 weeks of drug intervention, the level of ALT and AST was detected, and the protein and mRNA expressions of MMP-9 and TIMP-1 in liver and kidney were detected by Western Blot and realtime-PCR. Results Compared with the model group, the level of AST in Qishui group was significantly decreased(P<0.01), while the level of ALT was decreased(P>0.05). The protein and mRNA expression of MMP-9 in liver was significantly increased(P<0.01), while the protein expression of TIMP-1 was significantly decreased(P<0.01), and mRNA was decreased(P<0.05). The protein and mRNA expression of MMP-9 in kidney was increased(P<0.05), while the protein and mRNA expression of TIMP-1 was decreased(P<0.05). Conclusion kk-ay mice have liver injury in the later stage of diabetic nephropathy, and Qishui decoction has protective effects on liver and kidney injury in kk-ay mice.
