摘要:
目的 建立宽筋藤药材的超高效液相色谱(ultra performance liquid chromatography,UPLC)特征图谱,为宽筋藤药材的整体质量分析和评价提供参考依据。方法 采用Waters Acquity C18色谱柱(2.1 mm×50 mm,1.7 μm),流动相0.1%甲酸乙腈溶液(A)-0.1%甲酸水溶液(B),梯度洗脱,流速为0.5 mL/min,柱温为30 ℃,检测波长为320 nm。采用聚类分析和特征图谱相似度分析对16批不同地区的宽筋藤商品药材UPLC特征图谱进行评价。结果 建立了宽筋藤药材的UPLC特征指纹图谱共有模式,共标定了35个共有峰。聚类分析将16批宽筋藤商品药材分为4类,特征图谱经国家药典规定的相似度计算软件计算,整体相似度良好。结论 本研究建立的宽筋藤药材特征图谱的特征性和专属性强,方法准确、可靠、重复性好,可用于宽筋藤药材的整体质量控制和评价。
Abstract:
Objective To establish the ultra performance liquid chromatography(UPLC)specific chromatogram of caulis tinosporae sinenisis, in order to provide reference basis for overall quality analysis and evaluation of caulis tinosporae sinenisis. Methods Analysis was performed on the Waters Acquity C18 column(2.1 mm×50 mm, 1.7 μm), the acetonitrile with 0.1% formic acid(A)-0.1% formic acid(B)was adopted as mobile phase in gradient elution mode, and monitored under 320 nm wavelength. The flow rate was set at 0.5 mL/min and the column temperature was set at 30 ℃. Sixteen batches of caulis tinosporae sinenisis medicinal material samples collected from different areas were analyzed by hierarchical clustering analysis(HCA)and similarity analysis(SA)to evaluate the UPLC specific chromatogram. Results Thirty-five common peaks were selected to establish the UPLC specific chromatogram of caulis tinosporae sinenisis. Sixteen batches of medicinal material samples were classified as four clusters by HCA and a good overall similarity was calculated by the overall similarity software specified in the national pharmacopeia. Conclusions The established method for specific chromatogram by UPLC with better characteristic and specificity was accurate, reliable and repeatable, which could be applied to the overall quality control and evaluation of caulis tinosporae sinenisis.
